He was a consensus three-star recruit and committed to play college football at the University of Miami.

At Miami, Njoku played in 39 games over three seasons, catching 144 passes for 1,618 yards and 18 touchdowns. He was a first-team All-ACC selection in 2016.

Njoku was drafted by the Browns in the first round (29th overall) of the 2017 NFL Draft. He played in 16 games as a rookie, catching 56 passes for 639 yards and four touchdowns. In 2018, Njoku recorded 59 receptions for 639 yards and four touchdowns in 14 games before suffering a season-ending wrist injury.

Njoku missed the entire 2019 season after requesting a trade from the Browns. However, he returned to the team in 2020 and had a breakout season, catching 41 passes for 475 yards and four touchdowns. In 2021, Njoku signed a four-year, $54 million contract extension with the Browns.

Njoku is a versatile tight end who can line up in a variety of positions. He is a good blocker and receiver, and he is a threat to score from anywhere on the field. He is also a team player who is known for his work ethic and positive attitude.

Njoku was signed by the Cleveland Browns on June 15, 2017, to a four-year, $9.52 million contract with full guarantees and a $5.06 million signing bonus.

Njoku made his NFL debut against the Pittsburgh Steelers on September 10 in a 21-18 loss. His two receptions for a total of 20 yards. He had three receptions for 27 yards and his first NFL score in the Week 2 loss to the Baltimore Ravens, which ended 24-10.

He had 32 receptions for 386 yards and four touchdowns by the time his first season was up. Njoku started the 2018 season as the tight end for the Browns. He participated in all 16 contests, making 14 starts. His stats included 56 receptions for 639 yards and four touchdowns.

Njoku left the game against the New York Jets in Week 2 of the 2019 season after being knocked over and landing on his head.

Later that week, it was discovered that Njoku shattered his wrist during the same play. The injured reserve was activated on September 20, 2019.

On November 20, 2019, he was put back on injured reserve and started working out with the team once more. He started working on December 7, 2019.

The Browns chose to exercise Njoku's fifth-year option on April 27, 2020. On September 14, 2020, Njoku was placed on injured reserve due to a knee injury, one day after recording three receptions for 50 yards and a score against the Baltimore Ravens.

Njoku caught seven catches for 149 yards and a touchdown in Week 5 against the Los Angeles Chargers, with the score coming on a 71-yard catch and run. The final score was a thrilling 47-42 loss for the Browns.

On March 7, 2022, the Browns gave Njoku the franchise tag. Njoku extended his contract for four years and $56.75 million on May 27.

Njoku sustained a high ankle sprain in Week 7's game against the Baltimore Ravens. Later it was revealed that he wouldn't need surgery but would be out for two to five weeks.

The Cleveland Browns declared Njoku doubtful on September 30, 2023, before Week 4 because he had burns on his face and arms from a “freak accident” at home.
