Briefly played for the Philadelphia Eagles, Miami Dolphins, Baltimore Ravens, Cleveland Browns, and New York Jets before finding his footing with the San Francisco 49ers in 2016.

Became a breakout star in 2019, rushing for 919 yards and 14 touchdowns, including a memorable 43-yard touchdown run in the Super Bowl LIV.

Continued to be a key contributor for the 49ers, ranking among the league's fastest players.

Mostert signed with the Philadelphia Eagles as an undrafted free agent in 2015 after being selected without being selected in the NFL Draft. Mostert had a fantastic preseason, leading the NFL in yards from the line of scrimmage with his combined 351 yards from 157 rushing yards and 191 receiving yards.

In addition, he contributed 162 yards from five kick returns. Despite his best efforts, Mostert was finally cut on September 4, 2015, in order to free up cap space for the Eagles to reduce their roster to 53. However, two days later, he signed on to the Eagles practice squad.

Mostert was signed by the Miami Dolphins on September 14, 2015, after leaving the Eagles practice squad. In the Dolphins' Week 2 23–20 away loss to the Jacksonville Jaguars, he returned two kicks for 57 yards.

The Dolphins cut Mostert on October 13, 2015, with the intention of promoting him to the practice squad. Following the season-ending foot injury to third-string running back Lorenzo Taliaferro, Mostert was signed by the Baltimore Ravens on October 14, 2015.

The Cleveland Browns grabbed Mostert off waivers on December 16, 2015. For the last three games, he was the Browns' starting kick returner, returning 12 kicks for 309 yards.

 

Mostert was signed to the New York Jets practice squad on September 6, 2016, but he was cut loose six days later. Mostert signed a contract to join the practice squad of the Chicago Bears on September 13, 2016. He was added to the active roster on September 21.

In 2016, Mostert made two appearances for the Bears. On October 3, 2016, he was let go, and the following day, he joined on again with the practice squad. On November 24, 2016, the Bears released Mostert.

Mostert signed a contract to join the practice squad of the San Francisco 49ers on November 28, 2016. Dec. 31, 2016, saw his promotion to the active roster. Mostert participated in the 49ers' thrilling 25-23 loss to the Seattle Seahawks in the regular-season finale.

Mostert was placed on injured reserve on November 29, 2017. With six carries for 30 rushing yards and 83 return yards in 11 games, he concluded the 2017 campaign.

For the first five games of the 2018 season, Mostert was very rarely on the field. On the other hand, starting in Week 6, his role grew. In nine games with no starts, Mostert concluded the 2018 campaign with 34 carries for 261 yards and a score in addition to six receptions for 25 yards.

Mostert and the 49ers agreed to a three-year contract extension on March 15, 2019. Mostert asked for a trade from the 49ers on July 8, 2020. On July 27, after negotiating a revised contract with the 49ers, he withdrew his request.

Mostert got two carries for 20 yards in the season opener against the Detroit Lions before departing the game with an apparent knee injury. The team ultimately won 41–33 on the road.

Mostert inked a one-year deal with the Dolphins on March 17, 2022. He got 18 carries for 113 rushing yards and one rushing touchdown in the Jets game in Week 5, a 40–17 defeat.

Mostert and the Dolphins agreed to a two-year, $5.6 million contract agreement on March 15, 2023. Mostert had a major role in the Dolphins' 70-20 victory over the Denver Broncos in Week 3. He ran for 82 yards and three touchdowns and caught 60 passes for 60 yards and another score.

In the Miami Dolphins' 42–21 victory over the Carolina Panthers in Week 6, Mostert finished with 115 rushing yards, 17 receiving yards, and three touchdowns overall. For his efforts, he was named the AFC Offensive Player of the Week.

Mostert broke the record for the most touchdowns in a single regular season in Dolphins history in Week 15 against the New York Jets, scoring his 19th and 20th touchdowns of the campaign.
